Microdialysis technique


Microdialysis technique is able to continously monitor the biochemical changes in vivo in the extracellular fluid of the target tissue. The most important element of the microdialysis setup is the microdialysis probe. At the tip of the probe there is a semipermeable membrane. Depending on the cut-off value of the probe different exogenous and endogenous molecules can diffuse through the membrane during the perfusion process into the direction of the concentration gradiens. The protein unbound, pharmacologically active fraction of the test molecules can be measured by this technique at the end of the outlet tubing of the microdialysis probe where the dialysate samples are collected. For determination of the concentrations of the test drugs in the dialysate samples different, sensitive bioanalytical methods are developped. Our samples are usually analysed by LCMS/MS.
